Abstract

Island3 is a Cluster I1 bacteriophage that infects Mycobacterium smegmatis. Northwestern College students are performing systematic genetic screens to identify Island3 genes that are cytotoxic or protect host cells from infection by other phages (defense). We identified Island3’s gp36 as a cytotoxic gene. Now we are using a bacterial two-hybrid assay to identify a host protein(s) interaction partner. We hypothesize that host proteins that bind gp36 will reveal the mechanism by which gp36 confers cytotoxicity.

Bacterial 2-Hybrid Assay: Interactions of Phage Island3's Gene 36 and Mycobacterium smegmatis Gene Products
